摘要: Abstract The location and characteristics of atrial natriuretic peptide binding sites in the kidney toad, Bufo marinus, were determined. Specific 125 I-rANP observed on glomeruli blood vessels, but little if any was over regions corresponding to renal tubules. tissue sections and/or isolated membranes completely displaced presence 1 μM rat ANP, frog porcine C-type (membranes only); however, residual remained after incubation with NPR-C ligand, C-ANF, indicating two distinct sites. Electrophoresis cross-linked identified specific bands at approximately 70 140 kDa which correspond monomeric mass guanylate cyclase receptors, respectively. In addition, CNP stimulated a significant increase cGMP production rates membrane preparations, while C-ANF had no stimulatory effect. Two partial cDNA clones generated using primers based conserved vertebrate receptors showed high homology an (NPR-GC), This study provides evidence that B. marinus contains both NPR-GC glomerulus is potentially principal site ANP regulation kidneys.
