摘要: Following a single 50 mg dose of hydralazine (Apresoline) in 13 patients with impaired renal function, decrease glomerular filtration rate (GFR) was correlated an increase serum half-life (T1/2) the drug (r=-0.69; p less than 0.01). The T1/2 15.8 h one patient GFR 16 ml-min-1, as compared to 1.7-3.0 found previously healthy volunteers. In 49 on long-term antihypertensive treatment hydralazine, ratio between minimum steady-state concentration and daily (Cminss : Dose) increased decreased. This accumulation particularly evident 30 ml-min-1 (r=-0.63; 0.01; n=19). As excretion unchanged is generally regarded unimportant, slower elimination chronic failure probably caused by metabolic conversion. It found, however, that could easily have been underestimated, only 12.7% initial 200 ng-ml-1 urine be recovered after storage samples at room temperature for 24 h.
