Webcam-based flow cytometer using wide-field imaging for low cell number detection at high throughput

摘要: Here we describe a novel low-cost flow cytometer based on webcam capable of low cell number detection in large volume which may overcome the limitations current cytometry. Several key elements have been combined to yield both high throughput and sensitivity. The first element is commercially available 187 frames per second video capture at resolution 320 × 240 pixels. this design 1 W 450 nm laser module for area-excitation, with allows rapid interrogation field. final 2D flow-cell overcomes limitation hydrodynamic focusing higher sample wider This linear velocity target cells be lower than conventional “1D” flow-cells typically used cytometry similar volumetric rates. It also imaged full frame rate webcam. Using webcam-based wide-field imaging, it was confirmed that fluorescently tagged 5 μm polystyrene beads not practical due streaking from motion beads, did occur design. sensitivity then investigated using THP-1 human monocytes stained SYTO-9 florescent dye flow-cell. found detecting concentrations as mL rates 500 μL min−1 buffer blood. effectiveness concentration dependent: 100 84% were detected compared microscopy, 10 79% 59% detected.
