Effects of feeding bile acids and a bile acid sequestrant on hepatic bile acid composition in mice

摘要: An improved ultra performance liquid chromatography-tandem mass spectrometry (UPLC/MS/MS) method was established for the simultaneous analysis of various bile acids (BA) and applied to investigate liver BA content in C57BL/6 mice fed 1% cholic acid (CA), 0.3% deoxycholic (DCA), chenodeoxycholic (CDCA), lithocholic (LCA), 3% ursodeoxycholic (UDCA), or 2% cholestyramine (resin). Results indicate that have a remarkable ability maintain concentrations. The profiles mouse livers were similar between CA DCA feedings, as well CDCA LCA feedings. mRNA expression Cytochrome P450 7a1 (Cyp7a1) suppressed by all whereas Cyp7b1 only UDCA Gender differences composition observed after feeding CA, DCA, CDCA, LCA, but they not prominent UDCA. Sulfation found at 7-OH position, it increased more male than female mice. In contrast, sulfation taurolithocholic (TLCA) female-predominant, LCA. summary, present systematic study on metabolism will aid interpreting BA-mediated gene regulation hepatotoxicity.
