Callus growth and plant regeneration in diverse cultivars of cucumber (Cucumis sativus L.)

摘要: The growth and differentiation of callus tissues derived from cotyledons ten cultivars ofCucumis sativus L. were investigated. Cotyledonary explants all formed tissue on Murashige Skoog (MS) medium supplemented with 0.5 μM 2,4-dichlorophenoxyacetic acid 5 6-benzylaminopurine. Fresh weight the averaged 1 to 8 g per flask after five weeks culture. Shoot development was achieved in three cultivars, Hukchinju, Manchoonchoungjang Seoul, MS α-naphthaleneacetic Reducing 6-benzylaminopurine concentration 0.01 resulted root formation shoots transferred this medium. All gave same response tests formation, but shoot regeneration culture cucumber dependent genotype cultivar exhibiting best capability among genotypes examined. Examination mitotic metaphase regenerants revealed that tetraploid.
