A new lyophilized tachyzoite based ELISA to diagnose Besnoitia spp. infection in bovids and wild ruminants improves specificity.

摘要: Abstract Recent studies have reported that routinely used whole or soluble Besnoitia besnoiti tachyzoite (TZ) extract-based ELISAs potentially give rise to a high number of false-positive results, which may compromise control and the epidemiological bovine besnoitiosis. Thus, western blot (WB) has been recommended as confirmatory test. In present study, new ELISA test employs lyophilized tachyzoites for first time (BbSALUVET 2.0) was developed validated with cattle sera (n = 606) under worst-case scenario. False positive false negative, TZ BbSALUVET 1.0 reactors were overrepresented, WB reference One commercial (PrioCHECK Ab 2.0, extract) recently membrane-enriched (APure-BbELISA) also tested. The three showed AUC values (>0.9). However, best diagnostic performance corresponded 2.0 APure-BbELISA [(92% sensitivity (Se) 98% specificity (Sp)] followed by PrioCHECK (88% Se, Sp, 4.5% doubtful results). addition, wild ruminant sera, excellent (96% 97% 4% results) obtained again.
