Ginsenoside Rc, an active component of Panax ginseng, stimulates glucose uptake in C2C12 myotubes through an AMPK-dependent mechanism

摘要: Abstract Ethnopharmacological relevance Panax ginseng and its major component, ginsenosides, are widely used for the prevention of various disorders in oriental medicine. Aim study To evaluate effect ginsenoside Rc (Rc), one active constituents , on glucose uptake C2C12 myotubes. Results Treatment myotubes with significantly increased uptake. determine mechanism Rc-induced uptake, either insulin-dependent signaling or insulin-independent pathway activities were measured using western blot analysis. We showed that activated an AMPK pathway. However, had no components pathway, such as receptor substrates (IRS)-1 protein kinase B Akt (PKB/Akt). Moreover, we found treatment inhibitor abolished both p38 MAPK phosphorylation. This result implies activity is critical situated upstream MAPK. In addition, also activation induced by mediated reactive oxygen species (ROS) production, suggesting regulators AMPK- MAPK-mediated Conclusion Ginsenoside enhances inducing ROS generation, which leads to activation. Consequently, can be a potent natural anti-diabetic agent.
