摘要: Gua Lou Gui Zhi decoction (GLGZD), a traditional Chinese medicine consisting of different herbal medicines, has been used for centuries in the treatment muscular spasticity following stroke, epilepsy or spinal cord injury. However, precise mechanisms involved remain poorly understood. In present study, we investigated neuroprotective effects GLGZD on glutamate-induced apoptosis cultured BV-2 cells, as well underlying mechanisms. A 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was applied to assess viability cells. An Annexin V/propidium iodide (PI) utilized analyze cellular apoptosis. Mitochondrial membrane potential (MMP) evaluated by flow cytometry and laser scanning confocal microscopy. The gene protein expression apoptosis-related genes, Bcl-2 Bax, analyzed RT-PCR western blot analysis, respectively. Furthermore, cleaved caspase-3 detected immunofluorescence. Glutamate induced loss cell viability, which associated with an increase apoptotic rate, Bax/Bcl-2 ratio extracellular levels caspase-3. Treatment significantly reversed these phenotypes, its maximum protective observed at concentration 1,000 µg/ml. These results indicate that protects cells from cytotoxicity. may be ascribed anti-apoptotic activities, part, decrease expression, stability high mitochondrial potential.
