摘要: Abstract C-type lectin-like receptors (CLRs) are important pathogen pattern recognition molecules that recognize carbohydrate structures. However, the functions of these in fish keep less known. In this study, we characterized a novel CLR from teleost fish, Plecoglossus altivelis (ayu), tentatively named PaCD209L. The cDNA PaCD209L is 1464 nucleotides (nts) length, encoding polypeptide 281 amino acid residues with calculated molecular weight 31.5 kDa. Multiple alignment deduced sequences and other related CLRs revealed sequence had typical characteristics CLRs, but without Ca 2+ -binding sites. Sequence comparison phylogenetic tree analysis showed shared highest identity (44%) rainbow trout ( Oncorhynchus mykiss ) CD209 aE transcripts were detected all tissues examined, mainly expressed brain heart. Upon Vibrio anguillarum infection, upregulated tested monocytes/macrophages (MO/MΦ). We prepared recombinant (rPaCD209L) by prokaryotic expression raised antiserum against Western blot native was glycosylated, its protein significantly increased ayu MO/MΦ upon V. anguillarum infection. addition, rPaCD209L able to bind Gram-positive Gram-negative bacteria absence . After blocked anti-PaCD209L IgG, phagocytosis bacterial killing activity decreased. These results suggest plays an role regulation ayu.
