摘要: In embryos subjected to assisted reproductive techniques, epigenetic modifications may occur that can influence embryonic development and the establishment of pregnancy. horses, storage temperature during transport fresh before transfer is a major concern. The aim this study was, therefore, determine effects two temperatures (5 °C 20 °C) on equine embryos, collected at day seven after ovulation stored for 24 h, on: (i) morphological development; (ii) expression candidate genes associated with embryo growth development, maternal recognition pregnancy, methylation apoptosis, (iii) gene-specific global DNA methylation. Embryos (n = 80) were or eight assigned four groups: control (E7F, fresh); seven, h 5 (E5C); (E20C) (E8F, 24h time control). medium (EquiHold, holding medium, Minitube, Tiefenbach, Germany) from all treatment groups analyzed temperature, pH, lipid peroxidation (malondialdehyde; MDA) morphology, mRNA (immunohistochemistry methylation). size was larger (p < 0.01), whereas smaller 0.05) h. There no changes in pH MDA accumulation irrespective group. specific related (POU5F1, SOX2, NANOG), pregnancy (CYP19A1, PTGES2), (DNMT1, DNMT3A, DNMT3B) apoptosis (BAX) E5C E20C either up downregulated when compared controls (E7F E8F). immune 5mC 5hmC similar among groups. Percentage CpG islands lower ESR1, NANOG DNMT1 0.001) E8F (advanced stage). Therefore, our demonstrates first status days ovulation. Although results suggest some beneficial comparison °C, short-term storage, regardless modified gene involved compromise viability transfer.
