Searching of Standard Comet Assay Parameters for Detecting Lymphocyte DNA Damages Using Fourteen Different Test Conditions

摘要: The alkaline single cell gel electrophoresis technique (SCGE), also known as the comet assay, is an evaluation method for estimating DNA damage and genotoxicity. This assay used to measure single-strand breaks, alkali-labile sites cross-linking in individual cells. Although a simple, sensitive, rapid compare results of previous studies, it often difficult interpret because variability among protocols. Standardisation validation could provide invaluable information identification hazardous substances risk assessment environmental occupational exposure. Further use standardised include monitoring effectiveness medical treatment investigation variation response that may reflect genetic or influences. In this study, was evaluated 4 parameters, including tail moment (TM), intensity (TI), olive (OTM) (DNAt) length. 6 blood samples were collected from genetically unrelated healthy volunteers, 7 different volume ranges (2.5 ml 2.5 µl, diluted with PBS RPMI) tested these parameters. intraclass correlation coefficients (ICCs) statistically significant all 14 test conditions (r=0.87, p<0.001 TM; r=0.86, TI; r=0.73, p<0.05 OTM r=0.87, DNAt). conclusion, determination using small sample volumes can contribute worthwhile knowledge future studies.
