摘要: The role of epidermal growth factor (EGF) and its receptors in human cancers was studied using 24 cell cultures including 15 squamous carcinoma (SCC) the skin, oral cavity, esophagus. EGF found to inhibit colony formation all SCC cells at doses that are mitogenic many other cells, keratinocytes dermal fibroblasts. This inhibitory effect on SCCs specific, because did not some cases slightly stimulated tumor such as adenocarcinomas stomach, cervix, breast sarcomas. amounts these were measured by immunoprecipitation labeled proteins with anti-EGF receptor polyclonal antibody binding assay membrane preparations 125I-EGF. Of 13 tested, except 3 esophageal showed higher levels than normal keratinocytes, which contain 1.5 X 10(5) sites/cell. In general, skin cavity had large order 10(6)/cell, whereas 10(5)/cell. Some about twice A431 cells. values for equilibrium dissociation constant (Kd) nM. sensitivity correlated well elevated level 12 lines, significance obtained when data excluded. present observations suggest play a development SCCs.
