摘要: The filamentous fungi Monascus spp. have been used in the production of food colorants and health remedies for more than 1000 years Asia. However, greater attention has given to safety products because they contain citrinin, which is harmful hepatic renal systems. citrinin biosynthetic gene cluster characterized Monasucs aurantiacus. ctnB encoding an oxidoreductase located between pksCT ctnA. In this study, a replacement vector (pCTNB-HPH) was constructed disrupt with hygromycin resistance as selection marker. linear transformed into M. aurantiacus using protoplast CaCl2/polyethylene glycol (PEG) method. Three ctnB-disrupted strains were obtained by homologous recombination. comparison parental strain, ΔctnB mutants barely produced citrinin. These data confirmed that directly involved biosynthesis. Moreover, yields pigments two disr...
