摘要: Aim: Nasal transient receptor potential vanilloid 1 (TRPV1) stimulation with capsaicin produces serous and mucinous secretion in the human nasal airway. The primary aim of this study was to examine topical effects various TRP ion channel agonists on symptoms specific mucins: mucin 5 subtype AC (MUC5AC) B (MUC5B). Methods: Healthy individuals were subjected challenges TRPV1 (capsaicin, olvanil anandamide), ankyrin (TRPA1) (cinnamaldehyde mustard oil) a melastatin 8 (TRPM8) agonist (menthol). Symptoms monitored, lavages analysed for MUC5AC MUC5B, i.e. mucins associated airway diseases. In separate groups healthy subjects, biopsies brush samples using immunohistochemistry RT-qPCR. Finally, calcium responses ciliary beat frequency measured isolated ciliated epithelial cells. Results: All induced pain or smart. Capsaicin, oil also produced rhinorrhea. Lavage fluids obtained after challenge indicated increased levels whereas unaffected. MUC5B immunoreactivities primarily localized submucosal glands peptidergic nerve fibres, respectively. Although trpv1 transcripts detected samples, functional could not be Conclusion: Agonists TRPA1 release airways vivo. These findings may relevance regard regulation production under physiological pathophysiological conditions. (Less)
