Association of cough hypersensitivity with tracheal TRPV1 activation and neurogenic inflammation in a novel guinea pig model of citric acid-induced chronic cough.

摘要: Objective This study was performed to establish a novel model of citric acid-induced chronic cough in guinea pigs and investigate the pathogenesis hypersensitivity. Methods Healthy conscious inhaled acid (0.4 M) for 3 minutes twice daily 25 days. Cough reactivity evaluated, substance P (SP) calcitonin gene-related peptide (CGRP) bronchoalveolar lavage fluid were detected, transient receptor potential cation channel subfamily V member 1 (TRPV1) protein expression trachea bronchus determined. Tracheal bronchial tissues examined TRPV1. Results Inhalation 0.4 M increased coughing time-dependent manner: peaked at 15 days reached lowest level accompanied by similar changes SP, CGRP, TRPV1 expression. mainly observed mucosal submucosal layer bronchi. The areas positivity bronchi acid-treated animals significantly larger than control group. Conclusions Repeated inhalation can be employed pigs. hypersensitivity this is related tracheal activation neurogenic inflammation.
