Efficient and Specific Modifications of the Drosophila Genome by Means of an Easy TALEN Strategy

摘要: Technology development has always been one of the forces driving breakthroughs in biomedical research. Since time Thomas Morgan, Drosophilists have, step by step, developed powerful genetic tools for manipulating and functionally dissecting Drosophila genome, but room improving these technologies developing new techniques is still large, especially today as biologists start to study systematically functional genomics different model organisms, including humans, a high-throughput manner. Here, we report, first Drosophila, rapid, easy, highly specific method modifying genome at very high efficiency means an improved transcription activator-like effector nuclease (TALEN) strategy. We took advantage recently “unit assembly” strategy assemble two pairs TALENs designed modify yellow gene (on sex chromosome) novel autosomal gene. The mRNAs were subsequently injected into embryos. From 31.2% F0 fertile flies, detected inheritable modification involving entire process from construction detection modifications can be accomplished within month. potential applications this TALEN-mediated are discussed.
