摘要: The main goal of cartilage repair is to create functional tissue by enhancing the in vitro conditions more physiological in vivo conditions. Chondrogenic growth factors play an important role in influencing homeostasis. Insulin‑like factor (IGF)‑1 and transforming (TGF)‑β1 affect expression collagen type II (Col2) glycosaminoglycans (GAGs) and, therefore, targeted use could make chondrogenic redifferentiation efficient. In present study, human chondrocytes were postmortally isolated from healthy articular cultivated as monolayer or 3D pellet cultures either under normoxia hypoxia stimulated with IGF‑1 and/or TGF‑β1 compare impact different factors. mRNA levels specific receptors (IGF1R, TGFBR1, TGFBR2) analyzed at time points. Moreover, gene rates type 1 and 2 observed over a period 5 weeks. Additionally, hyaline‑like Col2 protein sulphated GAG (sGAG) quantified. Stimulation resulted enhanced IGF1R TGFBR2 whereas TGFBR1 cultures. monolayer, differences reached significance. This effect was pronounced hypoxic culture cultures, increased amounts sGAGs after incubation validated. summary, constructing profile regarding be helpful for application engineering.
