Stimulatory effects of neopterin on hematopoiesis in vitro are mediated by activation of stromal cell function.

摘要: The pteridine neopterin (NP) was shown to be produced by monocytes and is known a useful marker of immunological activation, although, its biological activity still unclear. Recently, we found that intravenous administration NP increased the numbers blood leukocytes, granulocyte-macrophage progenitor cells (CFU-GM) in bone marrow spleens mice. In order elucidate mechanism whereby stimulates hematopoiesis, effects on hematopoietic stem cell proliferation differentiation vitro were studied using long-term culture (LTMC) system with cloned stromal line, MS-5. Adding LTMC total, CFU-GM colony-forming unit spleen (CFU-S). also number soft agar system, but it did not enhance colony formation when target semi-purified (T, B adherent cell-depleted cells) cultured this suggesting directly affect progenitors. Conditioned medium obtained from NP-treated had much greater colony-stimulating than untreated cells. Furthermore, treatment stimulated production IL-6 GM-CSF All these findings suggest activating function.
