Methods Placental cross-barrier feto-maternal relationship levels, maternal blood pressure, haemodymamic on uterine arteries were evaluated. The effect growth weight fetal tissues, perinatal outcome, was investigated. Main outcome measures Maternal viscosity, vascular indices ductus venosus, plasmatic levels sildenafil, embryo/fetal litter weights, perinatal/postnatal survival rates. Results Sildenafil citrate crossed placenta. its metabolite desmethyl-sildenafil, demonstrated a positive linear correlation treated animals versus controls; selective hypotensive without changes resistance noted days E8 E11 (embryonic day). lower pulsatility index venosus day E18 suggests overflow at end pregnancy. Effects offspring placental liver enlargement, increased gain second half pregnancy (irrespective enlargement) birth. Perinatal postnatal rates group remained unaltered. No haemodynamic evident animals. Conclusions In rats, appears have onset pregnancy, implying supply, weight, but no mortality.
