Prolonged inhibition of nitric oxide synthesis in pregnant rats: effects on blood pressure, fetal growth and litter size

摘要: In order to investigate the effect of chronic inhibition nitric oxide synthesis along pregnancy, pregnant rats were given drinking water alone (control group) or containing nonselective synthase inhibitor Nω-nitro-L-arginine methyl ester (L-NAME; 15 mg/day/rat equivalent 50 mg/kg/day; treated group), from postmating days 1 18 pregnancy. On 17 urinary volume, sodium and potassium excretion, urine protein concentration measured. Systolic blood pressure (SBP) was recorded daily. days 6, 11 pregnancy number sites implantation, embryos, litter size, fetal placenta weight determined. (mmHg) increased (p<0.001) on 2nd day administration L-NAME remained high throughout experiment. This treatment excretion volume (p<0.01), with changes in rate study. day 6 gestation group, implantation (0.14±0.10) significantly decreased (p<0.05) compared control group (1.45±0.58), but day 11 embryos similar both groups. By day 18 caused a substantial decrease (6.30±0.77 12.00±0.92 g), (3.17±0.22 4.74±0.21 g) size (7.95±0.59 11.95±0.45 fetus/litter; p<0.001).
