Cutaneous reactions induced by oxcarbazepine in Southern Han Chinese: incidence, features, risk factors and relation to HLA-B alleles.

摘要: Abstract Purpose Oxcarbazepine (OXC) is a promising alternative for patients who cannot tolerate carbamazepine. Recently, however, it has been reported that OXC-induced cutaneous adverse drug reactions (cADRs) are prevalent and may lead to discontinuation. Additionally, these thought be associated with HLA-B*1502. This study aims investigate the incidence, features risk factors of OXC-cADRs, explore their relation HLA-B alleles in Southern Han Chinese. Methods A prospective was performed which 252 new users were recruited. To examine association between OXC-cADRs alleles, 14 maculopapular eruption (MPE) cases, including 9 additional cases beyond this observation, genotyped by PCR-SSP sequencing. Thirty-five OXC-tolerant served as controls. Results Five (2.0%) developed an OXC-cADR, all mild MPE. History other AED allergy ( p =0.005, OR=121.23) non-AED =0.006, OR=59.92) significant multivariate logistic regression analysis. Only one patient OXC-MPE positive HLA-B*1502; frequency HLA-B*1502 did not differ significantly from Four HLA-B*1302 detected different general population southern Chinese =0.001, OR=7.83). Conclusions The incidence cADRs low, no severe occurred. Patients history more susceptible OXC-cADRs. No found. associations HLA warrant further studies.
