摘要: To understand the roles of Rho-kinase and myosin light chain kinase (MLCK) for contraction organization stress fibers, we treated cultured human foreskin fibroblasts with several MLCK, Rho-kinase, or calmodulin inhibitors analyzed F-actin in cells. Some cells were transfected green fluorescent protein (GFP)-labeled actin, effects also studied these living The Y-27632 HA1077 caused disassembly fibers focal adhesions central portion cell within 1 h. However, located periphery not severely affected by inhibitors. When washed fresh medium, gradually reformed, 3 h completely recovered. ML-7 KT5926 are specific MLCK disruption and/or shortening peripheral leaving relatively intact even though their number was reduced. W-5 W-7 gave essentially same results as inhibitors, but to lose spread morphology, indicating that play a major role keeping flattened state cell. fiber models reactivated, contracted before fibers. Thus our study shows there at least two different systems system is dependent more on activity than while depends MLCK.
