A simple method for obtaining peritoneal macrophages from chickens.

摘要: Abstract A simple technique for inducing the production of peritoneal exudate cells within 3 to 4 days in chickens is described. In each 5 6 week old chicken, 8 ml washed Sephadex particles suspension saline (3%) were injected intraperitoneally. Three later, harvested, washed, suspended Hank's solution, counted and adjusted one million per ml. well 4-chamber slide 1 cell was incubated at 37°C 30 min. Cultures then gently several times leaving a monolayer glass adherent cells. The phagocytic activity these examined using either latex or antibody-sensitized sheep erythrocytes. with solution (with excess particle/peritoneal ratio) h 37°C. From 99.5 100% found actively sensitized erythrocytes both. Adherent monolayers cultivated vitro weeks no detectable decrease their activity.
