Retroviral Transfer of Antisense Integrin α6 or α8 Sequences Results in Laminar Redistribution or Clonal Cell Death in Developing Brain

摘要: To assess the roles of two integrin alpha subunits (alpha6 and alpha8) in developing chicken optic tectum, progenitors were infected with retroviral vectors that contained marker gene lacZ plus antisense sequences from either alpha6 or alpha8 subunit cDNAs. On embryonic day 3 (E3), vector was injected into tectal ventricles embryos. E6, E7.5, E9, later, embryos killed, tecta dissected processed for histochemical detection lacZ-positive cells. The antisense-bearing cell clones (descendants a single progenitor) analyzed proliferation migration patterns compared lacZ-only vector-infected control clones. At both antisense-containing similar to controls. exhibited number reduction upper laminae (intermediate zone plate), at they ventricular as well. Integrin no difference total E9 but had net laminar redistribution more cells less plate. Our data show different integrins play during brain development: is essential specific laminae, survival tectum Also integrin-substrate interactions may suppress early programmed death premigratory migratory neuroblasts.
