摘要: Background: Patients with locally advanced rectal cancer are treated preoperative chemoradiotherapy followed by surgical resection. Despite similar clinical parameters (uT2-3, uN+) and standard therapy, patients’ prognoses differ widely. A possible prediction of prognosis through microRNAs as biomarkers out treatment-naive biopsies would allow individualized therapy options. Methods: Microarray analysis 45 microdissected from patients was performed to identify potential predict overall survival, disease-free cancer-specific distant-metastasis-free tumor regression grade, or nodal stage. Quantitative real-time polymerase chain reaction (qPCR) on an independent set 147 validate relevant miRNAs. Results: In the microarray screen, 14 were significantly correlated survival. Five included previous work. Finally, 19 miRNAs evaluated qPCR. miR-515-5p, miR-573, miR-579 miR-802 demonstrated significant correlation survival (p < 0.05). miR-573 also grade after chemoradiotherapy. miR-133b showed a miR-146b expression levels Conclusion: Specific can be used possibly stratify if validated in prospective study.
