摘要: In an effort to identify novel genes related the prognosis of gastric cancer, we performed gene expression profiling and found overexpressed levels human interferon-induced transmembrane protein 1 ( IFITM1 ). We validated cancer–specific up-regulation its association with cancer progression. also studied epigenetic regulation tumorigenesis-related functions. Expression was evaluated in various cells 35 patient tumor tissues by quantitative RT-PCR Western blot analyses. The results showed highly up-regulated cell lines tissues. Furthermore, IHC studies were on 151 tissues, a significant correlation revealed between higher Lauren's intestinal type P = 0.007) differentiated adenocarcinoma 0.025). Quantitative DNA methylation for 27 CpG sites regulatory region hypermethylation expressing low IFITM1. Methylation-dependent confirmed further vitro demethylation using 5-aza-2′-deoxycytidine luciferase assays. functional analysis silencing small-interfering RNA decreased migration invasiveness cells, whereas overexpression exhibited opposite results. this study, demonstrated regulated promoter role prognosis.
