Subchronic toxicological study of two artemisinin derivatives in dogs.

摘要: The objective of our study was to profile and compare the systematic changes between orally administered artesunate intramuscularly injected artemether at a low dose over 3-month period (92 consecutive days) in dogs. Intramuscular administration 6 mg kg-1 induced decreased red blood cell (RBC) count (anemia), concurrent extramedullary hematopoiesis spleen inhibition erythropoiesis bone marrow. We also observed prolonged QT interval neuropathic central nervous system, which demonstrated cortex motor neuron vulnerability, but no behavioral changes. Following treatment with artesunate, we heart rate, most likely due cardiac conduction system damage, as well deceased RBC count, However, contrast artemether, neurotoxicity not following artesunate. In addition, ultra-structural examination by transmission electron microscopy showed mitochondrial damage These findings spectrum toxic that result upon show doses these derivatives diverse toxicity profiles.
