摘要: Enterovirus 71 (EV71), one of the major causative agents Hand-Foot-Mouth Disease (HFMD), causes severe pandemics and hundreds deaths in Asia-Pacific region annually is an enormous public health threat. However, effective therapeutic antiviral drugs against EV71 are rare. Nucleoside analogues have been successfully used clinic for treatment various viral infections. We evaluated a total 27 nucleoside discovered that adenosine analogue NITD008, which has reported to be reagent specifically inhibits flaviviruses, effectively suppressed propagation different strains RD, 293T Vero cells with relatively high selectivity index. Triphosphorylated NITD008 (ppp-NITD008) functions as chain terminator directly inhibit RNA-dependent RNA polymerase activity EV71, it does not affect VPg uridylylation process. A significant synergistic anti-EV71 effect rupintrivir (AG7088) (a protease inhibitor) was documented, supporting potential combination therapy other inhibitors
