摘要: The existing cell surface markers used for sorting glioma stem cells (GSCs) have obvious limitations, such as vulnerability to the enzymatic digestion and time-consuming labeling procedure. Reduced nicotinamide adenine dinucleotide (NADH) a cellular metabolite with property of autofluorescence has potential be new biomarker GSCs. A method GSCs was established according properties NADH. Then, NADHhigh NADHlow subpopulations were sorted. stem-like evaluated by qRT-PCR, western blot analyses, limiting dilution assay, viability bioluminescence imaging, immunofluorescence analysis in vitro vivo. relationship between CD133+/CD15+ subpopulation also assessed. expressed higher stem-related genes, formed more tumor spheres, harbored stronger pluripotency tumorigenicity vivo, compared subpopulation. had similar stemness CD133+ or CD15+ GSCs, but three less overlaid each other. Also, invasive resistant chemotherapeutic drug temozolomide (TMZ) than cells. In addition, NADH might an appropriate marker sort cancer (CSCs) other types, breast colon cancer. Our findings demonstrate that intracellular is non-labeling, sensitive maker isolating even CSCs.
