Comprehensive two-dimensional liquid chromatography with ultraviolet, evaporative light scattering and mass spectrometric detection of triacylglycerols in corn oil.

摘要: An improved comprehensive two-dimensional (LC × LC) HPLC system for the analysis of triacylglycerols was developed. In first-dimension, a Ag(I)-coated cation exchanger (250 mm 2.1 mm, 5 ?m) employed with gradient from 100% MeOH to 6% MeCN in at 20 ?L/min. Using 10-way valve two switching loops, 1 min sections first-dimension were introduced second-dimension consisting 30 4.6 C18 (1.8 column an isocratic mobile phase methanol?methyl tert-butyl ether (70:30) 3.0 mL/min. As solvent stronger than solvent, focusing took place, leading better separations previously reported analyses which hexane main constituent eluent. Compounds differing by 2 their partition number baseline separated second-dimension. Detection place UV 210 nm, evaporative light scattering and (+)-atmospheric pressure chemical ionisation-MS latter giving best results. Corn oil investigated 44 compounds could be detected: 34 (TAGs), 8 oxygenated TAGs, TAGs containing trans double bond. Data manipulation allowed construction contour plots automated calculation first- retention times peak areas. Quantitative results are compared fatty acid methyl ester analysis, literature data.
