Long-term Alterations in the Permeability of the Blood-Testis Barrier Following a Single Intratesticular Injection of Dilute Aqueous Glycerol

摘要: The mechanism whereby glycerol exerts its antisper- matogenic action is not known. objective of this study was to determine, with the use (3Hjinulin and ('25l)albumin by in vivo vitro methods, whether effect altering permeability blood-testis barrier (BTB). Adult male rats re- ceived asingleintratesticular injection ofeitherglycerol (1 O% or 20%; treated) saline (control), 2, 4, 8, 26, 56 weeks after treatment, either (3H)inulin ('Ijalbumin administered infusion directly testicular tissues. Radioactivity measured tissue, rete testis fluid, seminiferous tu- bular fluid. Following administration, uptake tubules, tubule fluid significantly greaterinthetreatedthaninthecontroltestes at all times treatment. Radiolabeled inulin, injected into isolated testesoradded tomedium inwhichisolatedtubuleswere incubated, accumulated higher levels compartment treated than control Rete from testes, collected micropuncture following efferent duct ligation, contained about 5- 13-fold more radioactivity controls. 50 jCj of('Thljalbumin intothe jugular vein, greater epklidymal tissues animals. In experiments, significant differences between controlwere maintained duringthe period of2-56 followingglyceroltreatment.The results suggest that a single intratesticular dilute solution alters BTB macromolecules such as inulin albumin,and theeffect appearstobe permanent.
