Expression of Calcyclin in Human Melanoma Cell Lines Correlates with Metastatic Behavior in Nude Mice

摘要: Since our aim was to isolate and identify new progression markers of human cutaneous melanoma, we applied the differential hybridization technique, in which compared gene expression two subsequent stages this progression. Tumors nude mice arising after transplantation serial passage vivo either horizontally early vertically growing part or advanced a primary melanoma same patient were used for assay. This resulted isolation number complementary DNA clones that differentially expressed. Based on marked difference expression, one them, designated pMW1, chosen further characterization appeared be coding calcyclin, cell cycle-regulated protein, belonging family small calcium-binding proteins. Calcyclin elevated high-metastatic lines low-metastatic ones. Immunoprecipitation calcyclin showed at RNA level is also reflected protein level. These findings show related metastasis emphasize role proteins neoplastic as reported mouse homologue other members family.
