Evaluation of un-methylated DNA enrichment in sequencing of African swine fever virus complete genome.

摘要: African swine fever is a febrile hemorrhagic disease that caused by the virus (ASFV) and lethal for domestic pigs wild boar. ASFV also infects soft ticks of genus Ornithodoros, some species which can act as vector ASFV. Whole genome sequencing challenge because, due to size difference host versus viral genome, higher proportion DNA fragments renders poorly efficient. A novel approach enrichment, based on separation methylated un-methylated DNA, has been reported but without an evaluation its efficacy. In this study, efficiency enrichment protocol was evaluated pig tick samples infected As expected, fewer reads corresponding were found in fraction compared fraction. However, coverage not improved untreated DNA. our hands, inefficient very limited samples. This process represents extra work cost significant improvement coverage. The cost/benefit ratio are discussed.
