摘要: To evaluate the effect of manidipine 10 mg on 24-hour ambulatory blood pressure (BP) and heart rate (HR) in very elderly hypertensive patients, 54 patients aged 76-89 years (mean age 81.8 years) with systolic (SBP) > 160 mmHg diastolic (DBP) 90 were studied. After a 4-week placebo washout period, randomized to receive or placebo, both administered once daily for 8 weeks. Patients checked after initial run-in phase every 4 weeks thereafter. At each visit casual BP HR measured. end period active treatment, noninvasive measurement (ABPM) was performed. Manidipine significantly lowered sitting standing SBP (P < 0.001) DBP at trough level. ABPM showed significant decrease values 0.001), daytime night-time 0.005). In addition, confirmed consistent antihypertensive activity throughout dosing interval, without circadian profile. The trough/peak ratio 0.67 0.59 DBP. No statistically change observed. treatment well tolerated, there no serious side effects. conclusion, once-daily administration tolerated effective reducing as BP.
