Immunogenicity of insulin-producing cells derived from human umbilical cord mesenchymal stem cells

摘要: Mesenchymal stem cells (MSCs) have been considered as hypo-immunogenic and immunosuppressive. However, a thorough understanding of the immunological properties after MSC differentiation in vitro vivo has not reached. We asked whether it would be immunogenic or influenced by immune microenvironment transplantation. In different disease models, changes MSCs greatly varied, with contradicting results. order to clarify this, we used modified four-step induction method induce human umbilical cord (hUCMSCs) differentiate into insulin-producing (IPCs), investigate reactions transplantation diabetic mice. found that induced IPCs are hypo-immunogenic, lacking HLA-DR, CD40 CD80 expression. Of note, observed cell infiltration peritoneal cavity left kidney capsule local IPCs. This indicated hUCMSC-derived maintained vitro, but became transplanting host, possibly due thereafter enhancement infiltration.
