摘要: Parishin is a dominant active ingredient originating from Gastrodia elata Blume, and has good neuroprotective effects against brain disorders. In the present study, metabolic profile of parishin by in vitro vivo experiments was investigated using ultra-high performance liquid chromatography coupled with quadrupole-time flight mass spectrometry (UHPLC/Q-TOF MS) combined an automated MS(E) technique. By comparison reference compounds, accurate measurement, characteristic fragmentation patterns parent drug gastrodin relevant bio-transformation knowledge, 14 metabolites (seven hydrolyzates seven derivatives gastrodin) were detected identified rat plasma urine after intragastric administration parishin, including processes hydrolyzation, oxidation, sulfation glucuronidation. According to proposed pathways hydrolytic study plasma, it can be inferred that mainly functions as prodrug undergoes hydrolysis before being absorbed into blood. The hydrolyzate, gastrodin, involved further metabolism, which responsible for pharmacological activities parishin. conclusion, this work provides valuable information on metabolism rapid reliable UHPLC/Q-TOF MS method, could widely used investigation natural product.
