摘要: We have previously reported on the amplification and overexpression of MDM2 proto-oncogene in a subset malignant gliomas without TP53 mutation (G. Reifenberger et al. , Cancer Res., 53: 2736–2739, 1993). Here, we show that MDM4 ( MDMX ) gene located 1q32 is further target for gliomas. codes Mdm2-related protein can bind to p53 inhibits p53-mediated transcriptional transactivation. investigated series 208 (106 glioblastomas, 46 anaplastic gliomas, 56 low-grade gliomas) identified 5 tumors (4 glioblastomas 1 oligodendroglioma) with overexpression. Several other genes from were found be coamplified such as GAC1 five tumors, REN four RBBP5 three tumors. Additional analyses revealed carried neither mutations conserved regions nor gene. Taken together, our data indicate novel molecular mechanism by which small fraction human escapes p53-dependent growth control.
