Comparative genomic hybridization of human malignant gliomas reveals multiple amplification sites and nonrandom chromosomal gains and losses.

摘要: Nine human malignant gliomas (2 astrocytomas grade III and 7 glioblastomas) were analyzed using comparative genomic hybridization (CGH). In addition to the amplification of EGFR gene at 7p12 in 4 9 cases, six new sites mapped 1q32, 4q12, 7q21.1, 7q21.2-3, 12p, 22q12. Nonrandom chromosomal gains losses identified with overrepresentation chromosome underrepresentation 10 as most frequent events (1 2 astrocytomas, glioblastomas). Gain a part or whole 19 bands 9pter-23 22q13 detected each five cases. Loss band 17p13 gain 20 revealed three The validity CGH data was confirmed interphase cytogenetics YAC clones, painting tumor metaphase spreads, DNA fingerprinting. A comparison results banding analyses indicates that spreads accessible primary cell cultures may not represent clones predominant tissue.
