Maintenance of peritoneal macrophages in the steady state

摘要: Resident peritoneal macrophages (M phi) were labeled in situ by intraperitoneal (i.p.) injection of the green fluorescent cell tracking dye PKH-1. After immunofluorescence staining with M phi specific monoclonal antibodies (Mabs) and phycoerythrin (PE) second antibody, resident both red Mab label, while recruited only tag. These populations distinguished two-color flow cytometry. PKH-1 followed for 1-49 days mice that received no further treatment (steady state). Dye still detectable after 49 vivo, although their fluorescence intensity had decreased steadily over time. The decrease was limited to phi, as lymphocytes did not change. clearly separated from up 28 days. No number (dye labeled) observed 1-28 data indicate replaced blood monocytes steady state.
