Functional differentiation of normal human neutrophils

摘要: In the past differentiation of human neutrophils has been defined by morphology, cytochemistry, or surface markers. our experiments we have sequenced various events that occur during functional normal neutrophil and also examined some properties in relationship to markers biochemical events. Granulocytes were obtained from bone marrow blood hematologically individuals. Cells separated into different stages maturation their physical using counterflow centrifugal elutriation density gradient separation. Three cell fractions enriched for either immature myeloid cells, band neutrophils, segmented neutrophils. Since not entirely pure, methodologies assays chosen allowed cytologic evaluation capacity each type. The criteria used classify included both morphology light microscopy DNA labeling with tritiated thymidine. Various neutrophilic studied: Fc receptors, complement receptors (CR1, CR3), phagocytosis live dead opsonized Staphylococcus aureus, microbial killing S NBT dye reduction after cellular stimulation endotoxin, chemotaxis. Our results indicate appear a distinct order. sequence appears be following: receptors----immune phagocytosis----complement receptors----oxygen-independent killing----oxygen-dependent killing----chemotaxis.
