Alzheimer paired helical filaments: immunochemical identification of polypeptides.

摘要: Antisera to isolated Alzheimer neurofibrillary tangles (ANT) of paired helical filaments (PHF) were raised in rabbits. These anti-PHF sera immunolabeled both ANT sections hippocampus and which extracted with sodium dodecyl sulfate (SDS). The immunostaining tissue was removed by absorption the serum small amounts PHF also 40-fold amount a fraction prepared identically from normal brain; neurofilament brain microtubule preparations used at same concentration as control did not eliminate tangle staining. Furthermore, staining glial or actin myosin filaments. No labeling neurofilaments axons cerebellar basket fibers observed non-neurologic brain. On paper blots SDS-polyacrylamide gels reacted neither polypeptides nor major polypeptides. However, immunoblots revealed several polypeptide bands 45,000–70,000 molecular weight (MW) region, material on top gel diffuse high MW region. abolished its low areas SDS polyacrylamide but results indicate that (1) antigen(s) recognized is inherent PHF, (2) this polypeptide(s) least partly soluble SDS, (3) occurs associated microtubules, neurofilaments, actin, myosin.
