Concatemer chain reaction: A taq DNA polymerase-mediated mechanism for generating long tandemly repetitive DNA sequences

摘要: The concatemer chain reaction (CCR) uses Taq DNA polymerase to synthesize double- or single-stranded concatemers whose length and yield can be controlled by varying the number of thermal cycling steps. Although reactions which occur in CCR are slower more complex than (PCR), practical application technique is simple. less expensive, faster, easier conventional methods for producing gives greatly improved yields. templates used may be: (i) double-stranded produced ligation, (ii) from previous CCRs, (iii) oligonucleotides consisting one copy sense strand repeat a complementary but overlapping antisense strand. Different molar ratios lengths (masses) two strands helix obtained. We have both as targets RNA hybridization. Applications this technology discussed, including use hybridization probes applications such run-on transcription analysis repetitive sequences.
