摘要: The selective interaction of neutrophils with E-selectin and eosinophils P-selectin has been previously reported, but the relevance selectin site density fluid shear not studied in detail. We have developed a new approach to examine these interactions cell suspensions that integrates an on-line cone-plate viscometer flow cytometer. find both use glycoprotein ligand-1 form stable conjugates Chinese hamster ovary transfectants, preferential adhesion eosinophils. Further, difference between is magnified at expression levels below ∼20 sites/μm 2 , range likely be relevant endothelial conditions associated eosinophilia. unique behavior retained over rates ranging from 100 1500/s low shear. Results parallel-plate chamber assays suggest eosinophil reflects enhanced efficiency initial PSGL-1 bond formation rather than ability mediate rolling longer duration on low-density substrates. These differences may account part for increase accumulation allergic diseases.
