Regulation of Expression of MSG1 Melanocyte-Specific Nuclear Protein in Human Melanocytes and Melanoma Cells

摘要: MSG1 is a nuclear protein and possible transcriptional transactivator that expressed strongly in melanocytes but very weakly, if at all, most nonmelanocytic cells or adult mouse tissues. This strong expression of cultured normal human epidermal was found to be dependent on both endothelin-1 FGF-2. The phorbol ester TPA could substituted for endothelin-1. mRNA transcripts were rapidly induced by either TPA. However, FGF-2 had no effects the level, suggesting its contribution translational and/or posttranslational level(s). (as well as transcripts) melanoma cells, which produce an autocrine growth factor. Melanoma derived from primary tumors tyrosinase-positive metastatic after treatment, while tyrosinase-negative did not. TPA-induced correlated with TPA-dependent activation promoter sequence, indicating regulation. In vivo, detected nevocytic nevus confined pigmented region, showed cell-level heterogeneity These results demonstrate regulated transcriptionally posttranscriptionally local factors cellular status differentiation.
