摘要: Abstract Monoclonal antibodies (MAbs) of predefined specificity have been generated by utilizing a synthetic peptide reflecting amino acid positions 10-17 the Hu-rasT24 gene product as immunogen. These MAbs, designated RAP-1 through RAP-5 (RA, ras; P, peptide), shown to react with ras p21. Since Hu-ras reactive determinants (positions 10-17) predicted be within tertiary structure p21 molecule, it was not unexpected that denaturation cell extracts or tissue sections Formalin glutaraldehyde enhanced binding RAP MAbs. When paraffin-embedded Formalin-fixed and avidin-biotin complex immunoperoxidase method were used, MAbs clearly defined expression in majority human colon mammary carcinomas. The all abnormal ducts lobules from fibroadenoma fibrocystic disease patients negative, normal colonic epithelia examined. findings reported here form basis for quantitative radioimmunoassays translational provide means evaluate individual cells tissues benign, "premalignant," malignant lesions.
