摘要: Abstract Introduction Recent reports have shown that the combined use of platelet-rich fibrin (PRF), an autologous matrix, and mineral trioxide aggregate (MTA) as root filling material is beneficial for endodontic management open apex. However, potential combination MTA PRF odontogenic inducer in human dental pulp cells (HDPCs) in vitro has not yet been studied. The purpose this study was to evaluate effect on odontoblastic maturation HDPCs. Methods HDPCs extracted from third molars were directly cultured with extract (PRFe). Odontoblastic differentiation evaluated by measuring alkaline phosphatase (ALP) activity, expression odontogenesis-related genes detected using reverse-transcription polymerase chain reaction or Western blot. Mineralization formation assessed alizarin red staining. Results treated PRFe significantly up-regulated dentin sialoprotein matrix protein-1 enhanced ALP activity mineralization compared those treatment alone. In addition, induced activation bone morphogenic proteins (BMP)/Smad, whereas LDN193189, protein inhibitor, attenuated sialophosphoprotein expression, treatment. Conclusions This shows a synergistic stimulation via modulation BMP/Smad signaling pathway.
