Effects of phosphodiesterase inhibitors caffeine and pentoxifylline on spontaneous and stimulus-induced acrosome reactions in human sperm

摘要: Objective To determine whether the phosphodiesterase inhibitors caffeine and pentoxifylline influence acrosome reaction in conditions which they are currently used as sperm movement enhancers. Design The frequency of occurring spontaneously capacitating media or induced by physiological (follicular fluid [FF]) artificial (ionophore A23187) stimuli was compared presence absence inhibitors. Setting Private hospital research laboratory. Patients, Participants Patients undergoing routine semen examination before vitro fertilization (no pathology detected) healthy donors. Interventions None. Main Outcome Measure Percentage acrosome-reacted determined with use fluorescein-labeled Pisum sativum agglutinin acrosomal stain. Results Caffeine alone augmented reaction, but this effect not observed alone. However, increased responsiveness to reaction-inducing stimuli, FF ionophore A23187. Conclusions promotion spontaneous may counteract benefits from application motility stimulant. On other hand, sensitization is expected contribute improvement fertilizing ability make drug a potential candidate for treatment anomalies.
