Radiation-induced epidermal growth factor receptor nuclear import is linked to activation of DNA-dependent protein kinase

摘要: Ionizing radiation, but not stimulation with epidermal growth factor (EGF), triggers EGF receptor (EGFR) import into the nucleus in a probably karyopherin α-linked manner. An increase nuclear EGFR is also observed after treatment H2O2, heat, or cisplatin. During, this process, proteins Ku70/80 and protein phosphatase 1 are transported nucleus. As consequence, an kinase activity of DNA-dependent (DNA-PK) increased formation DNA end-binding complexes containing DNA-PK, essential for repair DNA-strand breaks, occurred. Blockade by anti-EGFR monoclonal antibody C225 abolished radiation-induced activation inhibited repair, radiosensitivity treated cells. Our data implicate novel function during processes.
