摘要: We examined effects of the anti-epidermal growth factor receptor monoclonal antibody C225 on proliferation, cell cycle phase distribution, apoptosis, and radiosensitivity in squamous carcinoma (SCC) lines derived from head neck cancer patients. Exposure to culture inhibits SCC proliferation a time-dependent manner, degree inhibition, compared controls, ranges 20 75%. Flow cytometry analysis demonstrates that treatment induces accumulation cells G1, which is accompanied by 2-3-fold decrease percentage S phase. exposure also apoptosis populations, as demonstrated flow using dual stainings merocyanine 540 Hoechst 33342. Western blot indicates hypophosphorylated retinoblastoma protein increases expression p27KIP1. An increase Bax concurrent Bcl-2 are observed when exposed C225. Examination radiation response SCCs enhancement amplification radiation-induced apoptosis. These both single-dose fractionated experiments. represents promising growth-inhibitory agent can influence cellular neck.
