摘要: Tissue engineering of bone and oral mucosa have been extensively studied independently. The aim this study was to develop investigate a novel combination in single 3D vitro composite tissue mimicking the natural structure alveolar with an overlying mucosa. Rat osteosarcoma (ROS) cells were seeded into hydroxyapatite/tri-calcium phosphate scaffold constructs cultured spinner bioreactor for 3 months. An engineered fabricated by air/liquid interface culture immortalized OKF6/TERET-2 keratinocytes on collagen gel-embedded fibroblasts. EOM incorporated using adhesive further prior qualitative quantitative assessments. Presto Blue assay revealed that ROS remained vital throughout experiment. histological scanning electron microscope examinations showed proliferated densely populated construct. Micro computed tomography (micro-CT) increase closed porosity decrease open total at end period. Histological examination bone-oral model relatively differentiated parakeratinized epithelium, evenly distributed fibroblasts connective layer widely spread within scaffold. feasibility fabricating cell lines is demonstrated. Generating human ‘normal’ cell-based models characterization required optimize vivo applications.
